Key facts

  • Phase 2 tested 1, 3, 6 and 9 mg once weekly by injection under the skin, plus two escalation arms (6 to 9 mg and 3 to 6 to 9 mg).[1]
  • Phase 1 single doses ranged from 0.04 mg to 12 mg; the 12-week phase 1 used 1.2 to 12 mg weekly without escalation.[3][4]
  • Weight loss rose with dose: 9.5% at 1 mg up to 20.1% at 9 mg after 48 weeks.[2]
  • Phase 3 (ENLIGHTEN) tests four doses, listed only as Dose 1 to Dose 4 on ClinicalTrials.gov.[6]
  • This page reports trial protocols. It is not dosing advice, and eloralintide is not approved for any use.

Eloralintide Doses in the Phase 2 Trial

The 48-week phase 2 trial gave eloralintide once weekly at fixed doses of 1, 3, 6 or 9 mg, or escalated from 6 to 9 mg or from 3 to 6 to 9 mg, by subcutaneous injection.[1]

Phase 2 (n = 263), week 48. Weight change: efficacy estimand. Nausea: share of participants affected.

Lilly reports that GI side effects were lower with slower dose escalation and similar to placebo at 1 mg and 3 mg.[2] The 3 to 6 to 9 mg arm had less nausea (25%) than 6 mg started directly (64%).[1]

Eloralintide Doses in Phase 1

Phase 1 tested single doses from 0.04 mg to 12 mg in healthy adults, then once-weekly doses of 1.2, 3, 6 and 12 mg for 12 weeks without escalation.[3][4]

  • Single ascending doses: 0.04, 0.12, 0.4, 1.2, 4 and 12 mg, 6 people per dose. One 12 mg dose cut weight by 4.4% at week 4.[3]
  • Twelve-week multiple doses (n = 100): placebo-adjusted weight change of -2.6% at 1.2 mg, -8.9% at 3 mg, -8.5% at 6 mg and -11.3% at 12 mg.[4]
  • Exposure rose in proportion to dose, and the about-two-week half-life gave steady levels with weekly dosing.[4]

Eloralintide Dosing Schedule

Every eloralintide trial so far has used once-weekly subcutaneous injections, supported by a human half-life of about 310 to 366 hours.[3][4]

In the EloraTZP phase 2b, eloralintide started at 1 mg or 3 mg and rose to 6 mg or 9 mg, with combination arms escalating every 4 weeks.[5]

Phase 3 Doses

The ENLIGHTEN phase 3 trials compare four eloralintide doses with placebo, but ClinicalTrials.gov lists them only as Dose 1 to Dose 4, so the exact phase 3 doses are not public.[6]

Research Vial Strengths

DBR Labs supplies eloralintide for laboratory research in 5 mg, 10 mg and 20 mg lyophilised vials.

Vial strength describes the amount of powder in the vial. It is not a dose. Our products are for research use only and are not for human or veterinary use. See the vials.

Frequently Asked Questions

What doses of eloralintide were used in trials?

Phase 2 used 1, 3, 6 and 9 mg once weekly, plus escalation arms of 6 to 9 mg and 3 to 6 to 9 mg. Phase 1 tested single doses from 0.04 to 12 mg and 12 weeks of 1.2 to 12 mg weekly.

How often is eloralintide given?

Once a week by injection under the skin, in every trial so far. Its half-life of about two weeks supports weekly dosing.

What dose of eloralintide gave the most weight loss?

In phase 2, 9 mg weekly gave the largest mean loss at 48 weeks (20.1%), closely followed by the 6 to 9 mg escalation arm (19.9%).

Is there an approved eloralintide dose?

No. Eloralintide is not approved anywhere, so there is no approved dose. This page reports trial protocols only and is not medical advice.
